
低温驯化对西伯利亚蝗滞育卵过冷却点和自由水及糖原含量变化的影响

【目的】 西伯利亚蝗Aeropus sibiricus是高山草原牧草的重要害虫,一年繁殖一代,以卵在土壤中越冬,越冬经历高寒地区的低温,卵的耐低温能力直接影响越冬率。本研究旨在明确西伯利亚蝗卵越冬期间的耐低温特征,为进一步揭示其高寒适应能力和分布特点及发生量预测预报提供基础资料。【方法】 对越冬期滞育卵进行不同低温(10、5、0、﹣5、﹣10 ℃)驯化7 d,以25 ℃的滞育卵为对照组,测定不同低温驯化后滞育卵的过冷却点(Supercooling point, SCP)、自由水含量、糖原含量。【结果】 低温驯化对西伯利亚蝗滞育卵的SCP有显著影响,随驯化温度的下降SCP降低(P<0.05);滞育卵自由水含量随驯化温度的下降而减少(P<0.05);滞育卵糖原含量随着驯化温度的下降,呈先减少后上升再减少的趋势(P<0.05)。【结论】 西伯利亚蝗滞育卵随驯化温度的下降,其SCP显著降低,自由水含量和糖原含量显著减少,在越冬期通过降低SCP和减少自由水含量的策略抵御冬季低温,并调整体内糖原含量增强卵的耐低温能力。

[Aim] To
investigate the low temperature tolerance characteristics of overwintering Aeropus sibiricus eggs to reveal their
adaptability to alpine regions, distribution characteristics and provide basic
data for predicting the occurrence of this pest. [Methods] Diapause
eggs were exposed to different temperatures (10, 5, 0, ﹣5 and﹣10 ℃) for
7 d with diapausing eggs at 25 ℃ serving as the control group. The
supercooling point (SCP), free water content and glycogen content of eggs in
the treatment and control groups were then measured and compared. [Results] The
SCP, free water and glycogen content of eggs were all significantly lower at
lower temperatures (P<0.05).
Glycogen content first decreased during the early stage of temperature acclimation, increased during the middle stage, then
decreased again in the late stage. [Conclusion] Lower
temperatures significantly decrease the SCP, free water content and glycogen
content, of diapausing eggs of A.
sibiricus. The strategy of reducing SCP, free water and glycogen, content
is used during the overwintering period, to resist winter low temperatures.
