【目的】 旨在分析西方蜜蜂Apis mellifera鞘氨醇-1-磷酸磷酸酶1(Sphingosine-1-phosphate phosphatase 1, AmSgpp1)基因的分子特征、保守基序、结构域和系统进化,并解析AmSgpp1的时空表达模式,为AmSGPP1的功能研究提供参考和基础。【方法】 通过PCR验证AmSgpp1的表达,相关软件分析AmSGPP1的分子特征、保守基序和系统进化等。RT-qPCR检测AmSgpp1在工蜂成虫中肠等7个组织中的相对表达量;进一步检测AmSgpp1在卵、3日龄(d)(幼虫)、5 d(幼虫)、7 d(预蛹)、9 d(预蛹)和11 d(蛹),以及工蜂成虫1、2、6、8、12 d中的相对表达量。【结果AmSgpp1在西方蜜蜂工蜂中真实表达,CDS含有1 302个核苷酸,编码433个氨基酸。AmSGPP1的分子质量约为49.53 kD,蛋白质脂溶系数为107.37,等电荷点为8.59,氨基酸平均亲水系数为0.321,可同时定位于质膜、细胞质和过氧化物酶体。AmSGPP1含有7个跨膜结构域,但不含信号肽。西方蜜蜂与其他8个物种的SGPP1蛋白均包含1个相同的PAP2_SPPase1结构域和10个相同的保守基序。AmSgpp1在西方蜜蜂工蜂成虫的7个组织中差异表达,在脂肪体和中肠组织的表达量最高,毒腺次之。AmSgpp1在西方蜜蜂的卵、幼虫期和蛹期差异表达,在卵中的表达量最高,9 d(预蛹)最低,其表达量随发育时间逐渐降低;AmSgpp1在工蜂1、2、6、8和12 d成虫体内差异表达,在12 d成虫中表达量最高,且显著高于其它时期(P < 0.05)。【结论】 AmSGPP1是疏水性的胞内跨膜蛋白;西方蜜蜂与其他物种的SGPP1具有较高的基序和结构域的保守性,以及氨基酸的同源性;AmSgpp1在西方蜜蜂的不同组织和发育阶段中差异表达。

英文摘要:

 [Aim]  To analyze the molecular characteristics, conserved motifs, structural domains and phylogeny of the sphingosine-1-phosphate phosphatase 1 (AmSgpp1) gene of the western honeybee (Apis mellifera), and analyze the spatio-temporal expression pattern of AmSgpp1. [Methods]  The expression of AmSgpp1 was verified by PCR, and its molecular characterization, conserved motifs and phylogeny were analyzed using the relevant software. RT-qPCR was used to detect the relative expression of AmSgpp1 in the antennae, brain, hypopharyngeal glands, midgut, venom glands, epidermis and fat body, of adult worker bees, in eggs, 3-day-old larvae, 5-day-old larvae, 7-day-old pre-pupae, 9-day-old pre-pupae and 11-day-old pupae, and in 1, 2, 6, 8 and 12-day-old adult worker bees. [Results]  AmSgpp1 is authentically expressed in worker bees. Its CDS contains 1 302 nucleotides, encoding 433 amino acids. AmSGPP1 has an approximate molecular weight of 49.53 kD, a lipid solubility coefficient of 107.37, an isoelectric point of 8.59, and an average hydrophilic coefficient of 0.321. It can be simultaneously localized to the plasma membrane, cytoplasm and peroxisomes. AmSGPP1 contains seven transmembrane structural domains but no signaling peptide. The SGPP1 proteins of the A. mellifera and eight other species all contain one identical PAP2_SPPase1 structural domain, and 10 identical conserved motifs. AmSgpp1 is differentially expressed in seven adult worker tissues, with the highest expression in the fat body and midgut tissues, followed by the venom glands. AmSgpp1 is also differentially expressed in the egg, larvae, and pupal stages, with the highest expression in eggs and the lowest in 9-day-old pre-pupae. However, in adult worker bees, significantly higher expression was recorded in 12-day-old adults than in younger adults (P < 0.05). [Conclusion]  AmSGPP1 is a hydrophobic, intracellular, transmembrane protein. A. mellifera SGPP1 exhibits high conservation of motifs, domains, and amino acid sequence homology with the SGPP1 of other species. AmSgpp1 is differentially expressed across various tissues and developmental stages in A. mellifera.
